PENENTUAN KINETIKA ENZIM SELULASE DAN APLIKASINYA TERHADAP TANDAN KOSONG KELAPA SAWIT (TKKS) YANG DIPRETREATMENT MENGGUNAKAN IONIC LIQUIDS
This study utilizes TKKS, which is rich in cellulose and has the potential to be processed into glucose. The conversion of cellulose into glucose is carried out through enzymatic hydrolysis. The kinetics of hydrolysis is determined using the Michaelis-Menten equation and transformed into the Lineweaver-Burk method to obtain Vmax and Km parameters. The research findings show that enzyme activity is directly proportional to substrate concentration. The enzyme activity at a substrate concentration of 75 mg/mL is 0.2285 U/mL, while at 150 mg/mL it is 0.3872 U/mL. The enzymatic kinetics results in a Vmax of 1.34 (µmol/mL)/minute and a Km value of 368 µmol/mL.minute. The high Km value indicates that the enzyme's affinity is less effective. The maximum rate of cellulase enzyme in converting CMC substrate to product is 1.34 µmol/mL CMC per minute.
