PENENTUAN SIFAT ANTIOKSIDAN DAN ANTIBAKTERI DARI EKOENZIM CAMPURAN LIMBAH KULIT BUAH DAN KULIT RIMPANG

Organic waste is the largest contributing component of the total waste that ends up in landfills. This waste accumulation causes various problems, including being a source of various diseases and a cause of soil and water pollution. The production of eco-enzymes is an effective and efficient solution for managing organic waste by converting it into a multi-purpose liquid through a fermentation process. Eco-enzymes are a type of organic compound produced by the fermentation of fresh waste, such as vegetables, fruit peels, and rhizome peels, which have the potential as natural antioxidant and antibacterial agents. This study aimed to evaluate the organoleptic properties, total acid content, total phenolic and flavonoid content, and biological activity of an eco-enzyme formulated from a mixture of fruit peel and rhizome peel waste. Antioxidant activity was tested using the DPPH method and is expressed as the IC50 value, while antibacterial activity against S. aureus was tested using the disc diffusion method. Organoleptic testing showed that the panelists liked the aroma of the eco enzyme with an acceptance level of 72.67%, which falls into the 'like' category. The panelists tended to like the color of the eco-enzyme with an acceptance level of 78%, approaching the 'extremely like' category. The performance of the eco enzyme received a panelist acceptance rate of 82%, placing it in the 'very much like' category. The pH decrease from 5 to 3.2 after a four-month fermentation period indicated that the fermentation process was successful. The produced eco-enzyme also tested positive for acetic acid and lactic acid. The total acid content of the eco enzyme was 4.28%, with a total phenolic content of 2.22 mg GAE/g and a total flavonoid content of 0.758 mg QE/g. The eco-enzyme exhibited antioxidant activity with an IC50 value of 27223.14±5015.44 ppm, categorizing it as a weak antioxidant. In contrast, ascorbic acid as the standard had an IC50 value of 13.24±4.82 ppm, categorizing it as a very strong antioxidant. The antibacterial activity against Staphylococcus aureus showed an average inhibition zone diameter of 6.67±1.04 mm at 2686.57µg/disc, while chloramphenicol as the positive control showed an average inhibition zone diameter of 19.00±1.32 mm at 25 µg/disc. Therefore, this eco-enzyme formulation has very limited potential as either an antioxidant or an antibacterial agent. Keyword : ecoenzyme, fruit peel, rhizome peel, antioxidant, antibacterial
