PERBANDINGAN AKTIVITAS ANTIOKSIDAN BENTUK EKSTRAK DAN NANOFIBER DARI EKSTRAK ETANOL BATANG KAYU KUNING (Arcangelisia flava)

Background. Arcangelisia flava (commonly known as yellow wood) has been traditionally utilized for its antioxidant properties and potential as an herbal remedy for various diseases, including infections and inflammation. However, studied focusing on the application of yellow wood extract in nanofiber membranes to enhance its active compounds are still limited. This study aims to compare the antioxidant activity of ethanol extracts from yellow wood stems in pure extract form and nanofiber form. Methods. An experimental study was conducted in vitro to test the antioxidant activity of the extract and its nanofiber form. The extraction of A. flava stem was performed through maceration, while nanofibers were created via electrospinning. Total phenolic content was measured using the Folin-Ciocalteu method, and flavonoid content with the quercetin method at concentrations of 0, 20, 40, 60, and 80 ppm. Antioxidant activity was analyzed using the DPPH (2,2-diphenyl-1-picrylhydrazyl) method at concentrations of 2, 5, 10, 15, 20, and 25 ppm, and results were interpreted by IC50 values. Results. The total phenol content in the ethanol extract of Arcangelisia flava was 19,31 ± 0,014 mg GAE/g, while in nanofibers it increased to 36.49 mg GAE/g. Flavonoid content in the ethanol extract was 12,63 ± 0,178 mg QE/g, whereas in nanofibers it reached 142,55 mg ± 0,066 QE/g. The DPPH test showed that the ethanol extract had an IC50 value of 63.01 ppm, indicating strong antioxidant activity, while the nanofibers demonstrated a lower IC50 value of 49.12 ppm, signifying even stronger antioxidant activity. Conclusion. Nanofibers containing Arcangelisia flava ethanol extract exhibited stronger antioxidant activity compared to the pure extract, reinforcing the potential application of nanotechnology to enhance the efficacy of active compounds in therapeutic use.
