UJI KADAR TOTAL FLAVONOID DAN SAPONIN PADA BIJI BUAH GAMBAS (Luffa acutangula) SEBAGAI FITOESTROGEN UNTUK POTENSI TERAPI MENOPAUSE

Menopause leads to a decline in estrogen levels, which can cause various physiological and psychological disturbances. Although hormone therapy may be beneficial, it carries the risk of serious adverse effects, thereby necessitating safer alternative options. Luffa acutangula (gambas), a member of the Cucurbitaceae family, is known to contain secondary metabolites such as flavonoids and saponins; however, studies on the bioactive constituents of its seeds remain limited. This study aimed to determine the flavonoid, saponin, and phytoestrogen content in the ethanol extract of Luffa acutangula seeds using UV-Vis spectrophotometry. This research was conducted as an in vitro experimental laboratory study. Seed samples were extracted using the maceration method with 96% ethanol. Phytochemical screening was performed for preliminary identification of active compounds and showed positive results for flavonoids and terpenoids, whereas saponins were not confirmed in the extract and therefore were not subjected to quantitative analysis. Quantification of flavonoids and phytoestrogens was carried out using UV-Vis spectrophotometry with quercetin and genistein standards at concentrations of 20, 40, 60, 80, and 100 ppm. Absorbance values of the standards and samples were analyzed using calibration curves to determine the total compound content in the extract. The results showed that the ethanol extract of Luffa acutangula seeds contained flavonoids at 9.369 ± 0.061 mg QE/g. Phytoestrogen quantification using genistein as the standard demonstrated a concentration of 0.769 ± 0.003 mg QE/g, categorized as weak phytoestrogen activity. In conclusion, the ethanol extract of Luffa acutangula seeds contains flavonoids with potential for further investigation regarding their biological activities in herbal drug development, while saponins were not confirmed in the extract, and the detected phytoestrogen content was low.
