PERBANDINGAN KADAR DAN KEMURNIAN PROTEIN DAUN KENIKIR (COSMOS CAUDATUS) YANG DIENDAPKAN DENGAN AMONIUM SULFAT PADA BERBAGAI KONSENTRASI
Kenikir leaves (Cosmos caudatus) contain various bioactive compounds and have a relatively high total protein content compared to other plant leaves. Ammonium sulfate precipitation is one of the common methods applied for protein isolation. However, there are currently no available data comparing the protein content and purity of kenikir leaves proteins precipitated with different concentrations of ammonium sulfate. Therefore, this study was conducted as a pilot study. This in vitro experimental research examined kenikir leaves precipitated with ammonium sulfate at concentrations of 45%, 65%, 85%, and 95%. The precipitation process was performed as a stage of protein isolation, but the analysis in this study was performed on the supernatant fraction to explore the possible presence of low molecular weight proteins that remain soluble at certain salt saturations. Protein content was determined based on absorbance at 280 nm, while purity was evaluated by the A260/A280 ratio. The results showed that increasing ammonium sulfate concentration affected both the protein content and purity obtained with the highest protein content observed at 45% and the highest purity at 85%. Furthermore, hypothesis testing showed significant differences in the protein content and purity of kenikir leaves (proteins precipitated at different ammonium sulfate concentrations. Based on the balance between protein content and purity of kenikir leaves, the optimal ammonium sulfate concentration range was determined to be 65–85%
