Polycystic Ovary Syndrome (PCOS) is an endocrine disorder characterized by an imbalance of LH and FSH hormones, insulin resistance, and dyslipidemia, which increase the risk of metabolic complications. Purple sweet potato (Ipomoea batatas L. Poir) is known to contain anthocyanins and flavonoids with antioxidant properties that contribute to improving lipid profiles. This study aims to evaluate the effectiveness of purple sweet potato extract on lipid profiles in a PCOS rat model. In vivo experiment with a pre–post test design involving 30 female Wistar rats, divided into five groups. The PCOS model was induced by administering testosterone propionate and a high-fat diet for 42 days, followed by a single intraperitoneal dose of streptozotocin. The ovarian tissues were thoroughly rinsed, fixed in 10% formalin buffered with PBS at pH 7.4, processed using standard histological techniques including paraffin sectioning at a thickness of 5 µm and hematoxylin–eosin (HE) staining, and subsequently examined microscopically at 4× magnification. The treatment groups received metformin or purple sweet potato extract at doses of 200, 400, and 800 mg/kg body weight. Blood samples were collected from the ophthalmic vein to measure fasting blood glucose levels, and the data were analyzed using SPSS Statistics version 26. The administration of testosterone propionate combined with a high-fat diet for 42 days successfully induced ovarian features characteristic of PCOS, but did not produce significant changes in metabolic parameters. Mortality of the rats due to streptozotocin toxicity prevented the continuation of purple sweet potato extract treatment. Induction of testosterone propionate and a high-fat diet effectively induces PCOS reproductive abnormalities in mice.
