Thalassemia is a genetic blood disorder that requires lifelong management, including regular blood transfusions and iron chelation therapy. This condition affects the quality of life of children, which is strongly influenced by their parents’ level of knowledge in providing care. This study aimed to determine the relationship between parents’ level of knowledge and the quality of life of children with thalassemia at Dr. Mohammad Hoesin General Hospital, Palembang. The research employed an analytic observational cross-sectional design involving a total of 61 respondents, consisting of parents of children with thalassemia treated at the hospital. Data were collected using a thalassemia knowledge questionnaire to assess parental knowledge and the Pediatric Quality of Life Inventory (PedsQL) Parent Report 4.0 to measure the children’s quality of life. The relationship between variables was analyzed using the Chi-square test. Of the 61 respondents, 35 (57.4%) parents had good knowledge, while 26 (42.6%) had poor knowledge. The average quality of life score among children was 80.23, with 35 (57.4%) categorized as having a high quality of life and 26 (42.6%) as having a low quality of life. The analysis revealed a significant relationship between parental knowledge and the quality of life of children with thalassemia at Dr. Mohammad Hoesin General Hospital, Palembang (p = 0.040).
