EFEK EKSTRAK BAWANG PUTIH (Allium sativum L.) TERHADAP KERUSAKAN PARU TIKUS WISTAR JANTAN YANG DIINDUKSI ASAP ROKOK

Background: Cigarette smoke contains various free radicals that can damage lung tissue, characterized by alveolar lumen widening, alveolar wall thickening, and inflammatory cell infiltration. Natural antioxidants such as garlic extract (Allium sativum L.) are believed to neutralize free radicals and reduce such tissue damage. Methods: In vivo study was conducted using 30 male wistar rats divided into five groups: negative control (cigarette smoke and distilled water), positive control (cigarette smoke and vitamin C 50 mg/kgBW), garlic extract 50 mg/kgBW, garlic extract 100 mg/kgBW, and a combination of vitamin C and garlic extract 100 mg/kgBW. Cigarette smoke induction was performed for 21 days with exposure to 3 cigarettes per day. After treatment, the rats’ lungs were observed histopathologically for alveolar lumen widening, alveolar septal thickening, and inflammatory cell infiltration. The data were analyzed using normality tests, homogeneity tests, paired t-tests, independent t-tests, one-way ANOVA, and post hoc tests in SPSS version 24 to ensure that the data distribution met the requirements for parametric testing and to evaluate significant differences among treatment groups in determining the effectiveness of garlic extract against lung damage. Results: Lung damage data across all groups were homogeneous. Administration of garlic extract at a dose of 50 mg/kgBW reduced the mean alveolar lumen widening, alveolar septal thickening, and inflammatory cell infiltration in rats exposed to cigarette smoke. Vitamin C produced the greatest improvement compared to the other treatment groups. Conclusion: Vitamin C effectively improved lung damage in wistar rats induced by cigarette smoke based on parameters of alveolar lumen widening, septal thickening, and inflammatory cell infiltration compared to garlic extract. The effective dose of garlic extract for improving lung damage in cigarette smoke–induced wistar rats was 50 mg/kgBW and was characterized by a flat-dose effect on the variable of inflammatory cell infiltration.
