ANALISIS JARINGAN PROTEIN DAN SENYAWA HERBAL UNTUK MENENTUKAN TARGET TERAPI PADA INFARK MIOKARD YANG BERHUBUNGAN DENGAN MITOKONDRIA

Introduction: Myocardial infarction (MI) is a leading cause of global mortality with reperfusion therapy as its the gold standard. However, the following myocardial ischemia-reperfusion injury (MIRI) still lacks effective pharmacologic treatment. Mapping protein interaction networks may help identify herbal-based therapeutic targets related to mitochondria which holds central role in energy metabolism, oxidative stress regulation, and cell death. Methods: This exploratory study data-mined MI-related and mitochondrial proteins from several databases. Herbal compounds were identified from primary research articles retrieved through several electronic databases. Overlapping protein sets were analyzed using STITCH and STRING, then visualized in Cytoscape. Key proteins were defined based on cluster formation and top-ten rankings in four network centrality parameters. Results: The intersection of the three datasets revealed proteins that consistently appeared across all groups and were involved in cell-cycle regulation, apoptosis, autophagy and cell death, intracellular signaling, regulation of oxidative reactions, lipid metabolism, ubiquinone biosynthesis, and calcium homeostasis. TP53, AKT1, CYCS, CREB1, MAPK8, HSPA4, BECN1, MTOR, and GSK3B consistently ranked within the top 10 across all centrality parameters. Conclusion: This study identifies several key proteins implicated in MI pathogenesis that are mitochondrial-related and concurrently targeted by herbal compounds, exhibiting strong functional roles and connectivity within the protein interaction network.
