EFEKTIVITAS EKSTRAK UBI JALAR UNGU (Ipomoea Batatas L.) TERHADAP RESISTENSI INSULIN PADA MODEL TIKUS PCOS

Background: Polycystic Ovarian Syndrome (PCOS) is an endocrine disorder in women of reproductive age that is often accompanied by insulin resistance. Purple sweet potato (Ipomoea batatas L.) is known to contain anthocyanins and flavonoids that function as antidiabetics. This study aims to determine the effectiveness of purple sweet potato extract on insulin resistance in a PCOS mouse model based on blood glucose levels, insulin levels, and HOMA-IR values. Method: An in vivo experimental study with a pre–post test design was conducted using 30 female Wistar rats divided into 5 groups. The PCOS model was induced using testosterone propionate and high-fat diet (HFD) for 42 days, followed by a single intraperitoneal dose of streptozotocin. The treatment groups received either metformin or purple sweet potato extract at doses of 200 mg/kg BW, 400 mg/kg BW, and 800 mg/kg BW. Blood samples were collected from the ophthalmic vein to measure fasting blood glucose levels and insulin. Ovarium tissue is taken for histopathologic examination. Data analysis was performed using SPSS Statistics 24. Results: All treatment groups experienced a significant decrease in blood glucose, insulin, and HOMA-IR values. Administration of purple sweet potato extract at a dose of 800 mg/kgBW was more effective than the positive control group and other doses in reducing blood glucose, insulin, and HOMA-IR values. Conclusion: Purple sweet potato extract can significantly reduce fasting blood glucose levels, insulin levels, and HOMA-IR values in Wistar rats with insulin-resistant PCOS. The effective dose for reducing fasting blood glucose levels, insulin levels, and HOMA-IR values is 800 mg/kgBW. Keywords: High-Fat Diet, PCOS, Purple Sweet Potato (Ipomoea batatas L.)
