PENGARUH KONSENTRASI PELARUT TERHADAP AKTIVITAS ANTIOKSIDAN EKSTRAK ETANOL BATANG KAYU KUNING (Arcangelisia flava)
Background: Arcangelisia flava, commonly known as yellow wood, is an Indonesian medicinal plant containing bioactive compounds such as phenols and flavonoids, which act as natural antioxidants. The antioxidant activity of plant extracts is strongly influenced by the type and concentration of solvent used during extraction. Therefore, selecting the optimal solvent concentration is essential to obtain the maximum yield of active compounds. Methods: An in vitro experimental study was conducted using ethanol extracts at concentrations of 50%, 70%, and 96%, measured by UV-Vis spectrophotometry. The phenolic content was determined using the Folin–Ciocalteu method, flavonoid content using the AlCl₃ method, and antioxidant activity using the DPPH method to determine the IC₅₀ value. Results: The ethanol extracts at concentrations of 50%, 70%, and 96% showed total phenolic contents of 0.57 ± 0.0017, 0.57 ± 0.0006, and 0.66 ± 0.0005 g GAE/g, respectively. The total flavonoid contents were 1.24 ± 0.0000046, 1.27 ± 0.00047, and 1.24 ± 0.000005 g QE/g, respectively. The IC₅₀ values were 9.85 ppm, 9.36 ppm, and 9.63 ppm, respectively, indicating a very strong antioxidant activity category. Conclusion: The solvent concentration affected the total phenolic content, flavonoid content, and antioxidant activity of the ethanol extract of yellow wood (Arcangelisia flava). The 70% ethanol was the most effective solvent for extracting these bioactive compounds. Keywords: Arcangelisia flava, ethanol, phenol, flavonoid, antioxidant, DPPH, IC₅₀.
