PENGARUH KEPOLARAN PELARUT TERHADAP PENGHAMBATAN XANTIN OKSIDASE EKSTRAK ETANOL BATANG KAYU KUNING (Arcangelisia flava)
Background. Hyperuricemia is a condition characterized by increased uric acid levels due to excessive activity of the xanthine oxidase (XO) enzyme. Arcangelisia flava is known to contain secondary metabolites, such as flavonoids, alkanoids, and terpenoids, which have been reported to exhibit potential inhibitory effects on XO. This study aims to determine the effect of variations in ethanol solvent polarity (50%, 70%, and 96%) on the XO inhibitory capacity of A. flava stem extract. The use of solvents with different polarities is expected to yield varying levels of XO inhibitors, necessitating further investigation to identify the optimal ethanol concentration for extracting polar and semipolar bioactive compounds. Methods. This research employed an in vitro laboratory experimental design with a post-test design. Data collection was performed through the maceration of A. flava stems using three solvent variations which was 50%, 70%, and 96% ethanol. The XO inhibition assay was conducted in vitro using UV-Vis Spectrophotometry, where absorbance was measured at 293 nm and the data were processed to determine the percentage of inhibition and the IC50 value. Results. The study identifies the presence of alkaloids, flavonoids, terpenoids, and quinones in the ethanolic extracts of A. flava. Upon testing, the 70% ethanol extract was found to be a strong inhibitor, indicating that this concentration was the most effective and optimal for extracting bioactive XO inhibitory compounds. Consequently, the 70% ethanol extract of yellow wood stem (Arcangelisia flava) holds significant potential as a natural anti-hyperuricemia agent.
