PENENTUAN KADAR TOTAL FENOLIK, KADAR TOTAL FLAVONOID, AKTIVITAS ANTIOKSIDAN DAN TOKSISITAS TERHADAP EKSTRAK DAUN PUTAT INDIA (Barringtonia acutangula)

Barringtonia acutangula or indian oak is a type of tree that are found in many swamps and river banks in Sumatera Selatan region. Determination of total phenolic content, total flavonoid content, antioxidant activity and toxicity of indian oak (Barringtonia acutangula) leaves extract was carried out. Extraction process was carried out by using increasing polarity maceration with n-hexane, ethyl acetate and methanol solvent. Determination of total phenolic content and total flavonoid content was conduct using Folin-Ciocalteu method and AlCl3 (aluminium chloride) method respectively. Antioxidant activity was conduct using DPPH (2,2-diphenyl-1-picrylhydrazyl) method. Toxicity was conduct using BSLT (Brine Shrimp Lethality Test) method. The result of this research showed that the total phenolic content of n-hexane, ethyl acetate and methanol from leaf extract of B. acutangula were 6,54 ± 0,20; 41,18 ± 0,13 and 240,66 ± 1,64 mg GAE/g respectively. The total flavonoid content of n-hexane, ethyl acetate and methanol from leaf extract of B. acutangula were 20,53 ± 0,24; 100,59 ± 0,52 and 17,99 ± 0,43 mg QE/g respectively. The antioxidant activity test showed that the n-hexane extract was not active with IC50 value were 1670,65 ± 156,45 mg/L. The ethyl acetate extract was weak with IC50 value were 265,22 ± 10,59 mg/L. The methanol extract was very strong with IC50 value were 19,59 ± 0,08 mg/L. Ascorbic acid was used as standard and the antioxidant activity was very strong with IC50 were 11,18 ± 0,29 mg/L. The toxicity test showed that the n-hexane extract was not toxic with LC50 were 1000,2 ± 69,65 mg/L. The ethyl acetate and the methanol extract were toxic with LC50 value were 395,42 ± 217,16 and 362,34 ± 218,66 mg/L respectively.
