SINTESIS NANOPARTIKEL PERAK (NPAg) MENGGUNAKAN EKSTRAK DAUN TUMBUHAN Antidesma ghaesembilla DAN UJI ANTIBAKTERI TERHADAP Escherichia coli dan Staphylococcus aureus

Antidesma ghaesembilla or what is known locally as Sendulang has potential as a bioreductor in the synthesis of silver nanoparticles because it contains secondary metabolite compounds, one of which is flavonoids. A. ghaesembilla leaves have never been reported or studied for use as a bioductor in the synthesis of silver nanoparticles. Therefore, this research aims to synthesize silver nanoparticles using A. ghaesembilla leaves as well as antibacterial tests against Escherichia coli and Staphylococcus aureus bacteria. The synthesis of nanoparticles began by using Antidesma ghaesembilla leaf extract which was reacted with 40 mL of 0.001 M AgNO3 solution at varying extract volumes of 3, 5 and 7 mL and varying synthesis temperatures of 50, 60 and 70 ℃ for 1 hour. The silver nanoparticles formed were proven using UV-Vis spectrophotometry which showed the appearance of a Surface Plasmonic Resonance (SPR) absorption peak at a wavelength of 400-450 nm. The optimal condition of silver nanoparticles is determined by the absorbance value at the SPR absorption wavelength. Next, silver nanoparticles under optimal conditions were characterized using XRD and FTIR instruments and tested for antibacterial activity. The results of phytochemical screening of the methanol extract of A. ghaesembilla leaves show that A. ghaesembilla leaves contain secondary metabolite compounds, namely alkaloids, flavonoids, phenolics and terpenoids. The formation of NPAg can be seen visually by changing the color of the solution, namely from colorless to brownish yellow. Optimization results were obtained at a volume of A. ghaesembilla leaf extract, namely 7 mL and a temperature of 70 ℃ with an SPR absorption peak at a wavelength of 441 nm with an absorbance value of 0.837. The results of characterization using 16.38 nm particles with a crystal structure formed in the form of Face Center Cubic (FCC). FTIR results show a decrease in the absorbance value of the -OH group from 0.5154 to 0.1061, which indicates that the -OH functional group plays a role in the reduction reaction and as a stabilizer of the synthesized silver nanoparticles. The yield of AgNPs crystals obtained was 2 grams. AgNPs antibacterial test results of A. ghaesembilla leaf extract at a concentration of 2000 ppm against E. coli and S. aureus bacteria. AgNPs was more effective in inhibiting S. aureus bacteria with a disc diameter of 10.7 mm compared to E. coli bacteria of 8.25 mm. Based on this research, it can be concluded that NPAg can be synthesized using A. ghasembilla leaves and has potential as an antibacterial source
