KARAKTERISASI LIPOSOM PHOTOSENSITIZER PEMBAWA EKSTRAK KASAR NANAS (Ananas comosus (L.) Merr) SEBAGAI AGEN TROMBOLITIK
Thrombosis is a condition that occurs due to an imbalance in the hemostasis system characterized by the formation of a blood clot (thrombus) in the blood vessel wall so that it can cause ischemic stroke disease. The anti-thrombotic drugs commonly used are thrombolytics. These drugs contain fibrinolytic enzymes such as protease enzymes. Pineapple (Ananas comosus (L.) Merr) is known to have an anti-thrombotic pharmacological effect because it contains proteolytic enzymes, namely bromelain enzymes. Liposomes are used as a drug delivery system (DDS) because of their ability to protect enzymes from degradation. This study aims to determine the type of phospholipid that can produce the best thrombolytic activity. The types of phospholipids used for the manufacture of these liposomes are phosphatidylcholine from soy lecithin and Lipoid S100®, both of which have different phosphatidylcholine purities, namely 65% and 94%. Based on the results of the thrombolytic activity test, the liposome formula using Lipoid S100® (Lipoid, Germany) had the best results with a liposome size of 599.2 nm; PDI of 0.516; zeta potential of -13.4 mV and spherical liposomes. The characterization results for F1 and F3 showed good %EE, photothermal efficiency of 49.75% and 25.53%, and thrombolytic activity values with 450 nm laser testing of 27.74% and 25.54%. The results of this study show good thrombolytic results, but further research is needed to improve particle dispersion and increase phycocyanin stability.
