PENGARUH INTERMITTENT FASTING TERHADAP GAMBARAN HISTOLOGI DAN DIAMETER PULAU LANGERHANS PADA MENCIT MODEL OBESITAS

Background: Obesity is a global health problem that contributes to metabolic disorders, one of which is through changes in the structure and function of the pancreas, particularly the islets of Langerhans. Intermittent fasting, especially time- restricted feeding, is known to have the potential to improve metabolic parameters and tissue structure, but its effect on histology and islet diameter in obesity has not been extensively studied. This study aims to determine the effect of intermittent fasting on histological features and Langerhans islet diameter in an obese mouse model. Methods: This study was an experimental analytical study with a post-test only with control group design. The subjects were male mice (Mus musculus) divided into four groups, namely normal control, obesity control, obesity group with TRF intermittent fasting using high-fat feed, and obesity group with TRF intermittent fasting using standard feed. Pancreatic tissue preparations were stained with hematoxylin-eosin (HE). Histological assessment of Langerhans islets was performed using a damage scoring system, while the diameter of Langerhans islets was measured using ImageJ software. Data analysis was performed using statistical tests appropriate for the data distribution. Results: The results showed significant differences in the histological appearance of the islets of Langerhans between groups. The obese group showed an increase in islet damage scores compared to the normal control group. Administration of TRF- type intermittent fasting, both with a high-fat diet and a standard diet, significantly reduced islet damage scores. Conclusion: Time-restricted feeding intermittent fasting had a significant effect on improving the histological appearance of Langerhans islets in obese mice. Although changes in the diameter of Langerhans islets were found, this treatment did not show any statistically significant differences between treatment groups.
