Introduction: Oxidative stress is an imbalance between free radicals and endogenous antioxidants that plays a role in the pathogenesis of various degenerative diseases. Natural antioxidants from plants, particularly those containing phenolic compounds and flavonoids, have become potential alternatives for the prevention of oxidative stress. Kersen mistletoe leaves contain various secondary metabolite compounds with pharmacological activities, one of which can act as an antioxidant. This study aims to determine the antioxidant activity of the ethyl acetate extract from kersen mistletoe leaves. Methods: This study was an experimental in vitro study. The research sample used was kersen mistletoe leaves that were made into simplicia and extracted using the maceration method with ethyl acetate as the solvent. The extraction product underwent phytochemical testing and antioxidant assay, along with vitamin C solution as a positive control, using the DPPH method with a UV-Vis spectrophotometer. The percentage inhibition values were entered into a linear regression equation to obtain the IC₅₀ value. Results: The ethyl acetate extract of kersen mistletoe leaves contains flavonoid, alkaloid, tannin, and steroid compounds. The IC₅₀ value of the ethyl acetate extract of kersen mistletoe leaves was 10.77 ppm, while the IC50 value of vitamin C was 41.86 ppm. Conclusion: The ethyl acetate extract of kersen mistletoe leaves possesses very strong antioxidant activity. Keywords: Kersen Mistletoe Leaves, Ethyl Acetate Extract, Antioxidant
