KEANEKARAGAMAN KOMUNITAS BENTIK DENGAN MENGGUNAKAN METABARCODING eDNA SEDIMEN DI PERAIRAN PULAU ENGGANO, BENGKULU
Traditional monitoring has time and resource limitations, and can potentially harm organisms and destroy habitats. Anthropogenic is an issue to improve understanding of marine biodiversity. Therefore, alternative methods that are more effective, efficient, and environmentally friendly such as metabarcoding are needed. This study aims to analyze the sediment eDNA metabarcoding method and analyze the diversity of benthic communities in Enggano Island waters. COI gene identification using universal primers forward mlCOIintF 5'-GGWACWGGWTGAACWGTWTAYCCYCC-3' and reverse dgHCO2198 5'-TAAACTTCAGGGTGACCAAARAAYCA-3'. Sediment eDNA extraction in the six samples there are only two samples with an optimal purity value in sample S2L (Pulau Dua) 1.8 with a DNA concentration value of 7.1 nanograms/microliters and in sample S4L (Banjarsari Village) purity value of 1.95 with a DNA concentration of 13.2 nanograms/microliters. The diversity index value on Pulau Dua (0.9) is low and Banjarsari Village (1.2) is medium. The evenness index value of Pulau Dua (0.84) has high evenness criteria, while Banjarsari Village 0.57 criteria for medium evenness. The dominance index value of Pulau Dua (0.54) and Banjarsari Village (0.55) have medium criteria. Pulau Dua has low community stability diversity. While Banjarsari Village has moderate community stability diversity.
