EFEKTIVITAS EKSTRAK DAUN KERSEN (MUNTINGIA CALABURA.L) TERHADAP KADAR TNF-α PADA TIKUS WISTAR MODEL HIPERURISEMIA
Background: Hyperuricemia is a condition associated with increased systemic inflammatory responses, characterized by elevated levels of tumor necrosis factor alpha (TNF-α) and uric acid. Allopurinol is one of the first-line treatments for hyperuricemia. Cherry leaf (Muntingia calabura L.) is known to contain flavonoids with anti-inflammatory and antihyperuricemic activities. Methods: An in vivo study with a pre- and post-test control group design was conducted using 30 male Wistar rats induced to develop hyperuricemia using potassium oxonate. The rats were divided into five groups: negative control (0.5% Na-CMC), positive control (Allopurinol), and cherry leaf extract at doses of 100 mg/kgBW, 200 mg/kgBW, and 400 mg/kgBB. Drug and extract treatments were administered 1 hour after induction for 14 days. After treatment, blood samples were collected to measure uric acid and TNF-α levels using spectrophotometry and ELISA. Data were analyzed using tests of normality and homogeneity, paired t-test, Wilcoxon test, independent t-test, Mann Whitney test, Welch ANOVA, Kruskal Wallis test, Dunn post hoc test, and Games-Howell post hoc test using SPSS version 27 Result: Inflammatory responses among the Wistar rats were non-homogeneously distributed across all groups. Administration of cherry leaf extract at a dose of 400 mg/kgBW significantly reduced uric acid and TNF-α levels. Allopurinol demonstrated the most pronounced anti-inflammatory effect compared to other treatment groups in hyperuricemic rats. Conclusion: Allopurinol effectively reduced inflammation in potassium oxonate induced hyperuricemic Wistar rats based on TNF-α and uric acid parameters compared with cherry leaf extract. The effective dose of cherry leaf extract in reducing TNF-α and uric acid levels in hyperuricemic rat models was 400 mg/kgBW.
