PENGARUH MATERNAL SEPARATION TERHADAP PERILAKU KECEMASAN PADA MENCIT PUTIH (Mus musculus L.) GALUR SWISS WEBSTER JANTAN REMAJA

Background: Maternal Separation (MS) is a condition in which the mother and offspring are separated during the early stages of life, a critical period for the formation of physiological and psychological systems. This separation may induce stress that affects the Hypothalamus–Pituitary–Adrenal (HPA) axis, increases corticosterone levels, and alters brain structures such as the amygdala and hippocampus, which play key roles in anxiety regulation. Early-life stress can lead to long-term emotional disturbances. This study aimed to evaluate the effect of MS on anxiety behavior in mice during childhood and adolescence stages.Methods: This experimental study used a post-test only control group design with male Swiss Webster white mice as subjects. The treatment group was separated from their mothers for 4 hours daily until day 21, while the control group remained with their mothers. Anxiety behavior was assessed at 22 days (childhood phase) and 35 days (adolescent phase) using the Elevated Plus Maze (EPM). The parameters observed were the frequency and duration of entries into open and closed arms. Data were analyzed using Independent t-test and Paired t-test. Results: The results showed that the MS group exhibited decreased duration and frequency of entries into the open arms at 22 days compared to the control group, indicating increased anxiety behavior. However, at 35 days, the mice demonstrated greater exploration of the open arms and reduced anxiety compared to their earlier age, suggesting an adaptive process or partial recovery of stress regulation functions along with neural maturation. Conclusion: Maternal Separation for 4 hours per day increases anxiety behavior in juvenile mice, but this effect diminishes during adolescence due to neurophysiological adaptation mechanisms.
