UJI ANTINEFROLITHIASIS FRAKSI ETANOL DAUN KERSEN (Muntingia calabura L.) TERHADAP TIKUS PUTIH JANTAN YANG DIINDUKSI ETILEN GLIKOL DAN AMONIOUM KLORIDA

Kidney stone disease (antinefrolithiasis) of calcium stone type is one of the health problems among Indonesian people. The use of diuretic drugs as a treatment for kidney stones can be replaced with natural ingredients to reduce the risk of side effects. Kersen leaves (Muntingia calabura L.) contain a lot of flavanes, flavones, flavanones, flavonols, and chalcones which are included in the flavonoid group and are efficacious as diuretics. This study aims to determine the effect of ethyl acetate fraction of kersen leaves (EAFKL) as a kidney stone remover by examining urine pH and urine crystalluria, measuring blood serum creatinine and blood urea nitrogen (BUN) levels, measuring kidney calcium levels, measuring kidney organ index, macroscopic and histopathological observations of the kidneys. Test animals were divided into 7 groups, including normal group (NaCMC 0.5%), positive group (Batugin Elixir® 791 mg/kgBW), negative group (ethylene glycol 0.75% and ammonium chloride 1%), and 4 treatment groups (doses 60 mg/kgBW; 120 mg/kgBW; 180 mg/kgBW; 240 mg/kgBW). The results of urine pH and urine crystalluria examination showed that induction of ethylene glycol 0.75% and ammonium chloride 1% was able to produce calcium oxalate crystals in the kidneys. EAFKL doses of 60 mg/kgBW; 120 mg/kgBW; 180 mg/kgBW; 240 mg/kgBW are able to reduce blood serum creatinine and BUN levels, reduce kidney calcium levels, and reduce the index of the kidney organs of test animals that have been given induction. FEADK dose of 240 mg/kgBB has the highest effectiveness in the decay of kidney stones with an average value close to the normal group, so that the administration of a dose of 240 mg/kgBB provides higher effectiveness than the positive control group (p>0.05). Macroscopic results and organ histopathology showed good organ shape and a reduced amount of cell necrosis, glomerular atrophy, and crystal deposition. This indicates that the ethyl acetate fraction of kersen leaves has an activity in dissolving kidney stones with an effective dose (ED50) of 223,16 mg/kgBW.
