PRODUKSI ENZIM PROTEASE DARI Bacillus licheniformis TS-8 PADA MEDIA TEPUNG BULU AYAM DAN PEMURNIAN ENZIM MENGGUNAKAN KROMATOGRAFI PENUKAR ION
Protease is an enzyme capable of breaking peptide bonds in proteins into peptides and amino acids. Thermophilic bacteria from B. licheniformis are known as one of the microorganisms that produce protease capable of degrading keratin with high activity and under extreme conditions. In this study, protease enzyme production and purification were carried out from Bacillus licheniformis TS-8 isolated from the Tanjung Sakti Lahat Hot Spring and grown on chicken feather waste media. The crude extract obtained from the chicken feather media was concentrated using ultrafiltration and purified using ion exchange chromatography. Enzyme activity was determined using the Bergmeyer and Grab methods with Folin-Ciocalteu reagent, and protein content was determined using the Bradford method with Bradford reagent. The results of this study showed that crude enzyme extracts using chicken feather waste media had an enzyme activity of 0.7143 U/mL and a specific activity of 2.1587 U/mg with a protein concentration of 0.3309 mg/mL. The concentration results showed that the protein concentration increased approximately 2-fold, but the specific activity approximately the same. Protease enzyme activity was detected in NaCl elution with a concentration range of 0.1-1M, where the highest enzyme activity was found at a concentration of 0.2M, amounting to 4.3459 U/mL with protein content of 0.204 mg/mL and specific enzyme activity of 21.3034 U/mg with a purity level increased 13.66-fold.
