RESPON EKSPLAN DAUN DURIAN TERHADAP APLIKASI NAPHTHALENE ACETIC ACID PADA MEDIA WOODY PLANT MEDIUM

Durian is one of the leading commodities in South Sumatra, boasting high economic potential; however, its propagation is still constrained due to the difficulty in propagating it generatively. This research aims to determine the growth response of durian leaf explants to the application of the auxin NAA (Naphthalene Acetic Acid) in WPM (Woody Plant Medium). Research was conducted using a completely randomized design (CRD) with five levels of NAA concentration: N0 = 0 ppm, N1 = 1 ppm, N2 = 2 ppm, N3 = 3 ppm, and N4 = 4 ppm. Each treatment unit consisted of 10 culture bottles, repeated three times. The results of this research showed that the N0 treatment produced a 33% callus growth rate with a 13% contamination rate, while the N1 treatment showed a 6.7% callus growth rate with a 36.7% contamination rate. N2 produced a 60% explant survival rate, a 36.7% callus growth rate, and a 43% contamination rate. In contrast, treatment N3 showed the highest contamination rate of 76.7% and a live explant percentage of 23%. Treatment N4 showed a live explant percentage of 100% and a callus growth percentage of 6.7%. During the 8-week observation period, the highest callus formation occurred at a concentration of 2 ppm. This research concluded that NAA concentrations in the range of 0–2 ppm were more optimal for inducing callus growth, while higher NAA concentrations tended to increase the contamination rate and inhibit callus formation.
