Ethanol extract of breadfruit (Artocarpus altilis) stem bark is known to contain flavonoid compounds suspected to have potential as an antidepressant agent. This study was conducted to determine the effect and optimal concentration of the ethanol extract of breandruit stem bark for depression treatment. The test animals were divided into six groups: normal group (Na-CMC), negative group (Na-CMC+ Chronic Unpredictable Mild Stress (CUMS)), treatment groups I, II, and III (ethanol extract of breadfruit stem bark + Chronic Unpredictable Mild Stress (CUMS) at doses of 100 mg/kgBW, 200 mg/kgBW, and 400 mg/kgBW), and positive group (fluoxetine 20 mg/kgBW+CUMS). The test was carried out over 25 days, with body weight measurements taken on day 0, 5, 10, 15, and 20. Parameters observed in the study included changes in body weight, behavior, sucrose consumption, and braind histopathology. The result showed that the ethanol extract of breadfruit stem bark echibited significant antidepressant activity in chronic unpredictable mild stres induced test animals compared to the positive group (p>0,05). In general, the findings indicate that the ethanol extract of breadfruit stem bark has an antidepressant effect compared to the negative group.
