IDENTIFIKASI MORFOLOGI DAN UJI TOKSISITAS TERIPANG Holothuria fuscocinerea (Jaeger, 1833) TERHADAP Artemia salina (Linnaeus, 1758) DAN SEL KANKER PROSTAT
Sea cucumbers are a source of natural products containing triterpene glycoside saponin compounds with anticancer activity. The research conducted in January–July 2025 at the Research Center for Medicinal Raw Materials and Traditional Medicines, National Research and Innovation Agency, Bogor, Indonesia, aimed to identify species, analyze bioactive compounds, and test the bioactivity of lolosong sea cucumber extract from Bajo Village, Tilamota District, Boalemo Regency, Gorontalo as an anti-prostate cancer agent. The methods used included morphological and spicule identification, extraction using 70% ethanol, fractionation using n-hexane and 1-butanol, saponin quantification using the vanillin sulfate method, toxicity test of sea cucumber extracts and fractions against Artemia salina (Linnaeus, 1758) using the Brine Shrimp Lethality Test (BSLT) method, cytotoxic test of sea cucumber extracts and fractions against prostate cancer cells (PC-3) using the Microculture Tetrazolium Technique (MTT) Assay method, and compound analysis using Liquid Chromatography Mass Spectrometry (LC-MS). The results of the identification study showed that the observed lolosong specimen from Gorontalo was Holothuria fuscocinerea (Jaeger, 1833). Saponin quantification showed that the hexane fraction (3.995 ± 0.042 mg/mL) and butanol fraction (2.973 ± 0.032 mg/mL) had the highest saponin content. The butanol fraction showed the highest toxicity level in the BSLT test (LC50 57.48 µg/mL) and the PC-3 cell anticancer test (IC50 34.06 µg/mL). LC-MS analysis detected six putative compounds, namely Bivittoside A, Bivittoside B, Bivittoside C (=Bohadschioside B), Holothurinoside F, Holothurinoside F1 and Parvimoside A; all of which are non-sulfated saponins. This study shows the potential of the butanol fraction of H. fuscocinerea as a source of anticancer compounds.
