IDENTIFIKASI MORFOLOGI Bohadschia marmorata (Jaeger, 1833) DAN PENGUJIAN EKSTRAKNYA TERHADAP Artemia salina (Linnaeus, 1758) DAN SEL KANKER PROSTAT
Prostate cancer is one of the most common cancers affecting men and ranks fifth in Indonesia. Current treatments have adverse side effects and are costly, necessitating alternative medicines derived from natural sources. Marine organisms, particularly sea cucumbers, are known to contain triterpene glycosides with anticancer activity. This study aims to identify species, analyse bioactive compounds, and test the bioactivity of olo-olo sea cucumber extracts from Pesawaran Beach, Lampung, as an anti-prostate cancer agent. The study was conducted from January to July 2025 at the Research Centre for Pharmaceutical Ingredients and Traditional Medicine, National Research and Innovation Agency, Bogor, Indonesia. The research steps were morphological and spicule identification of the tested sea cucumbers, extraction of sea cucumber body wall with 70% ethanol, fractionation of the concentrated ethanol extract with n-hexane and 1-butanol, quantification of triterpene glycoside content using the vanillin sulphate method, dereplication of triterpene glycosides with LC-MS, testing the toxicity of the sea cucumber extract and fractions against Artemia salina (Linnaeus, 1758) with BSLT, and prostate cancer cells (PC-3) with the MTT Assay. Taxonomic identification results showed that olo-olo sea cucumber specimens from Lampung were Bohadschia marmorata (Jaeger, 1833) based on morphological and spicule analysis. The butanol fraction had the highest triterpene glycoside content (4.10 mg/mL) compared to the concentrated ethanol extract and other fractions. LC-MS analysis suggested putative compounds were holothurinoside I or I1, marmoratoside A or impatienside A, and DS-pervicoside B. The butanol fraction showed the most potential bioactivity against A. salina (LC50 11.60 µg/mL) and PC-3 cells (IC50 1.29 µg/mL) compared to other fractions. These results indicate the potential of the butanol fraction of B. marmorata as a source of natural anticancer compounds.
