EFEKTIVITAS MIKROBA DARI PEMBIAKAN BERSERI Colletotrichum sp., KITIN, DAN RIMPANG KENCUR TERHADAP PATOGEN ANTRAKNOSA CABAI

Chili plants in Indonesia face unique challenges in their cultivation, one of which is diseases affecting chili plants. Most common disease found in chili plants in Indonesia is anthracnose. Purpose of this study was to evaluate microbial effectiveness of serial cultivation of Colletotrichum sp., chitin, and kencur rhizomes against chili anthracnose pathogens. This research was conducted at Phytopathology Laboratory, Department of Plant Pests and Diseases, Faculty of Agriculture, Sriwijaya University. This research was conducted from July to November 2025. This research uses a split-plot design with main plot having 2 levels of enrichment material and subplot having 4 levels of serial cultivation cycles, with 3 replications. Main plot levels are Colletotrichum sp. + sugar enrichment material which code KeC and Colletotrichum sp. + chitin + sugar enrichment material which code KeCK. Subplot levels are 0, 1, 3, and 5 cultivation cycles. Observation variables in this study are colony growth inhibition, EC measurement, and microscopic observation of mycelium. In treatment M0, there was inhibition of growth of Colletotrichum sp. colonies, which was uniformly inhibited in each replication. In treatments S1-KeC, S3-KeC, and S5-KeC, there was inhibition, but in certain replications, there was no inhibition, and Colletotrichum sp. colonies originating from serial cultivation grew on media. In treatments S1-KeCK, S3-KeCK, and S5-KeCK, there was slight inhibition in certain replications, and Colletotrichum sp. colonies originating from serial cultivation also grew on media. Analysis of variance (ANOVA) of effect of serial cultivation treatments (KeC) on colony inhibition showed a tendency toward differences between treatments (p ANOVA = 0.054), and electrical conductivity (EC) value showed a significant effect between serial cultivation treatments (p ANOVA = 0.0003). Analysis of variance (ANOVA) of effect of serial cultivation treatments (KeCK) on colony inhibition showed a tendency toward differences between treatments (p ANOVA < 0.0001), and EC value showed a significant effect between serial cultivation treatments, as indicated by p ANOVA < 0.0001. Microscopic observation of treatments M0, S1-KeC, S3-KeC, and S5-KeC showed changes in mycelium. In each treatment, mycelial network appeared fragmented, and blue color of lactophenol cotton blue did not penetrate mycelium. Mycelium of Colletotrichum sp. forms numerous terminal or intercalary chlamydospores when grown on a glass slide. Antagonistic microbial cells derived from serial cultivation are able to inhibit colony growth, increase EC value of medium, and cause lysis of pathogenic fungus Colletotrichum sp. This research suggests adding pathogenic mycelium to treatment, maintaining sterility of tools and materials, and extending serial cultivation cycle.
