ALEL SPESIFIK IKAN NILA (Oreochromis niloticus) ASAL DANAU MANINJAU BERDASARKAN PENANDA POLIMORFISME NUKLEOTIDA TUNGGAL

SITI ANNISA TRI SUSANTI. Specific Allel of Nile Tilapia (Oreochromis niloticus.) from Maninjau Lake Based on Single Nucleotide Polymorphism (SNP) Marker (Supervised by MARINI WIJAYANTI). Tilapia (Oreochromis niloticus) is a fish native to the Nile River. Currently in Indonesia, there are many new strains of tilapia resulting from crossbreeding to produce tilapia with specific desired characteristics. Maninjau Lake is a lake used by the community for tilapia farming using floating net cage systems and also for capture fisheries. This study aims to determine the morphological, morphometric, and meristic characteristics, species composition based on specific alleles, and water quality of the tilapia habitat in Maninjau Lake, Agam Regency, West Sumatra. This study was conducted from November 2024 to January 2025. The research involved several stages, including sample collection, DNA extraction, DNA concentration measurement, electrophoresis, and SNP marker diagnostics. Morphological analysis of tilapia originating from Kaciak City, Tanjung Sani, and Maninjau Village showed that fish caught in the wild had a grayish body color with green patterns and gray fins with slightly red tips, while farmed fish had a black body color with a slight green or blue tint and black fins with red tips. Morphometry of wild cought tilapia show a total lenght of 11-12 cm and a body height of 1-8 cm, while farmed fish have a total lenght of 16.5-27.5 cm and a body height of 2-10.5 cm. The meristic measurements show relatively similar results. The allele composition of wild-caught and farmed fish resulted in a diagnostic allele frequency of 0,8 and an alternative allele frequency of 0.2 in wild-caught fish, while in farmed fish, the diagnostic allele frequency was 0.7 and the alternative allele frequency was 0.3. The results of water quality measurements at the three locations showed temperature ranging from 28-30℃, DO 3.8-7.7 mg L-1, ammonia 0.04-0.18 mg L-1, TDS 0.52-28 mg L-1, alkalinity 40-64 mg L-1 and pH 6,8-8,1.
