AKTIVITAS ANTIOKSIDAN DAN IDENTIFIKASI TENTATIF SENYAWA PADA FRAKSI KOLOM EKSTRAK ETANOL RIMPANG LEMPUYANG GAJAH (Zingiber zerumbet (L.) Roscoeex Sm.)
Oxidative stress caused by free radicals can cause significant disorders and contribute to the emergence of various chronic disease. The negative impact of these free radicals can be minimized by the use of antioxidants, such as the elephant lempuyang rhizome, which is often used as a medicinal plant and is believed to have antioxidant properties. This study aims to determine the antioxidant capacity of the elephant lempuyang rhizome and identify compounds that play a role in its antioxidant activity. Extraction was carried out using the maceration method and fraction separation was carried out using column chromatography. Each fraction was tested for screening antioxidant activity using the DPPH method and the selected fraction was identified using a Liquid Chromatography Mass Tandem Spectrometry (LCMS/MS) instrument. Fraction 3 demonstrated a lower IC₅₀ value of 123.44 µg/mL compared to the ethanolic extract of Zingiber zerumbet rhizome with an IC₅₀ value of 228.81 µg/mL, indicating that the fractionation process enhanced the antioxidant potential. Fraction separation increases the concentration of antioxidant compounds by separating inactive components, so that active compounds become more dominant. The results of the LCMS/MS analysis obtained luteolin compounds known to have antioxidant activity.
