AKTIVITAS ANTIOKSIDAN DAN IDENTIFIKASI TENTATIF SENYAWA PADA FRAKSI KOLOM EKSTRAK ETANOL UMBI BAWANG DAYAK (Eleutherine bulbosa (Mill.) Urb.)

Excessive exposure to free radicals can trigger oxidative stress, which plays a role in damaging various cellular components, including lipids, proteins, and genetic material, thereby increasing the risk of degenerative diseases. One approach to inhibiting this process is the utilization of antioxidant compounds derived from natural sources. Dayak onion bulbs (Eleutherine bulbosa (Mill.) Urb.) are traditional medicinal plants reported to contain secondary metabolites with potential antioxidant activity. This study aimed to evaluate the antioxidant activity of the ethanolic extract of dayak onion bulbs and to identify compounds suspected to contribute to this activity. Extraction was carried out using the maceration method, followed by compound separation using column chromatography. The antioxidant activity of each fraction was tested using the DPPH method, and the fraction with the highest activity, namely fraction 5.3 from the second column using an ethyl : ethanol eluent, was further analyzed using Liquid Chromatography Tandem Mass Spectrometry (LC-MS/MS). The results showed that antioxidant activity increased after the fractionation process, as indicated by a decrease in the IC₅₀ value from 385.919 µg/mL in the crude ethanolic extract to 56.436 µg/mL in fraction 5. LC-MS/MS analysis of the active subfraction tentatively revealed the presence of several secondary metabolites, with dithranol as the dominant compound based on the highest percentage area, as well as the identification of pterostilbene and eleutherol, which are known to be associated with the antioxidant activity of dayak onion bulbs.
