UJI AKTIVITAS ANTIOKSIDAN EKSTRAK ETANOL KULIT BATANG SUKUN (Artocarpus altilis (Park.) Fosberg) TERHADAP MENCIT (Mus musculus) YANG DIINDUKSI DEPRESI

The ethanol extract of breadfruit bark (Artocarpus altilis) contains active compounds such as flavonoids, phenolics, triterpenoids, tannins, and saponins, which exhibit antioxidant effects. This study aims to determine the characteristics, phytochemical profile, and the effect of the ethanol extract of breadfruit bark on antioxidant activity in mice subjected to chronic stress, using malondialdehyde (MDA) levels, catalase enzyme activity, and superdioxide dismutase (SOD) enzyme activity as parameters. The mice were divided into six groups: the normal group (0.5% NaCMC), the negative control group (0.5% NaCMC + chronic unpredictable mild stress (CUMS)), the positive control group (fluoxetine 20 mg/kg BW + chronic unpredictable mild stress (CUMS)), and treatment groups I, II, and III (ethanol extract of breadfruit bark at doses of 100, 200, and 400 mg/kg BW, respectively). The characterization results showed that the ethanol extract of breadfruit bark met the standards of the Indonesian Herbal Pharmacopoeia 2017. Antioxidant testing of the ethanol extract in depression-induced mice demonstrated decreased malondialdehyde (MDA) levels and increased catalase and superdioxide dismutase (SOD) enzyme activities compared to the positive control (p > 0.05). These results indicate that the ethanol extract of breadfruit bark exhibits antioxidant activity under chronic stress conditions compared to the positive control Keywords : Antioxidant, Artocarpus altilis, catalase, malondialdehyde, superdioxide dismutase
