UJI AKTIVITAS ANTIHIPERLIPIDEMIA EKSTRAK ETANOL DAUN KECIPIR (Psophocarpus tetragonolobus (L.) DC) PADA TIKUS WISTAR YANG DIINDUKSI DIET TINGGI LEMAK DAN FRUKTOSA
A study has been conducted to evaluate the antihyperlipidemic activity of the ethanolic extract of winged bean leaves (Psophocarpus tetragonolobus (L.) DC) in male Wistar rats induced by a high-fat and fructose diet. Male Wistar rats were divided into six groups: normal group, positive control (simvastatin 0.9 mg/Kg BW), negative control (0.5% NaCMC), and three treatment groups receiving ethanolic extract doses of 100 mg/Kg BW, 200 mg/Kg BW, and 400 mg/Kg BW. Total cholesterol levels were measured using the enzymatic CHOD-PAP method, HDL levels were determined using a Cholesterol HDL Analysis Kit with a photometer, while LDL levels were calculated indirectly. The reductions in total cholesterol and LDL levels as well as the increase in HDL levels in rats treated with ethanolic extract doses of 100, 200, and 400 mg/Kg BW showed significant differences compared to simvastatin (p < 0.05). Based on the percentage reduction of hyperlipidemic parameters, the effective dose (ED₅₀) of the ethanolic extract of winged bean leaves was determined to be 207 mg/Kg BW. Histopathological examination of the liver and heart tissues focused on necrosis and fatty degeneration. The results showed that the extract at a dose of 400 mg/Kg BW produced histopathological features comparable to simvastatin, with percentages of necrosis and fatty degeneration of 30% and 20%, respectively, while the positive control group showed 23% and 25%. These findings indicate that the ethanolic extract of winged bean leaves exhibits antihyperlipidemic activity and shows a significant difference compared to the negative control group (p < 0.05).
