AKTIVITAS ANTIOKSIDAN DAN IDENTIFIKASI TENTATIF SENYAWA PADA FRAKSI KOLOM EKSTRAK ETANOL RIMPANG BANGLE (Zingiber montanum)
Bangle rhizome (Zingiber montanum) is a medicinal plant with potential as a natural source of antioxidants owing to its phenolic and flavonoid contents. This study aimed to determine the total flavonoid content (TFC) and total phenolic content (TPC) of the ethanolic extract of a bangle rhizome, to evaluate the antioxidant activity of the selected fractions, and to identify the major antioxidant compounds in the active fraction. TFC was analyzed using the AlCl₃ method and expressed as mg QE/g, while TPC was determined using the Folin Ciocalteu method and expressed as mg GAE/g. Antioxidant activity was assessed using the DPPH method and evaluated based on IC₅₀ values. Extract fractionation was carried out by silica gel column chromatography using gradient elution based on solvent polarity, followed by thin-layer chromatography (TLC) monitoring to determine active fractions and subfractions. The subfraction exhibiting the highest antioxidant activity was further analyzed using Liquid Chromatography Tandem Mass Spectrometry (LC-MS/MS). The results showed that the ethanolic extract of a bangle rhizome had a TFC of 12.27 ± 0.121 mg QE/g and a TPC of 42.79 ± 0.030 mg GAE/g. The fractions exhibited very strong antioxidant activity with lower IC₅₀ values compared to the crude extract, indicating enhanced activity after fractionation. LC-MS/MS analysis of the active subfraction tentatively identified vanillic acid, zingerone, and curcumin, compounds known to possess antioxidant activity through free radical scavenging mechanisms. Based on these findings, the bangle rhizome has potential for development as a natural antioxidant source, and the fractionation process plays an important role in concentrating bioactive compounds responsible for antioxidant activity.
