AKTIVITAS ANTIOKSIDAN DAN IDENTFIKASI TENTATIF SENYAWA PADA FRAKSI KOLOM EKSTRAK ETANOL RIMPANG TEMU GIRING (Curcuma heyneana.)
Free radical molecules with unstable reactive properties due to the presence of unpaired electrons, so that they can form Reactive Oxygen Species (ROS) triggering oxidative stress. Oxidative stress plays a role in various health disorders so a source of antioxidants is needed to suppress it. The rhizome is one of Indonesia's medicinal plants with the potential as a source of antioxidants because it contains various secondary metabolite compounds, especially the terpenoid group. This study aims to evaluate the antioxidant activity of ethanol extract of rhizome, determine the fractions and subfractions with the highest antioxidant activity. Identify active compounds using the Liquid Chromatography Mass Spectrometry (LC-MS/ms) instrument. Extraction was carried out by the maceration method using ethanol solvent and the separation of fractions was carried out by column chromatography. Antioxidant activity was tested using the DPPH method and then assessed based on the percent inhibition and IC50 value. The separation of active fractions and subfractions was carried out through thin-layer chromatography (KLT) observation and DPPH qualitative tests. Selected subfractions were analyzed using LCMS/ms to identify active compounds. The results of the study obtained that antioxidant activity increased with fraction separation. The IC50 value of crude extract was 78.7954 μg/mL and fraction 4 was 45.5866 μg/mL. The results of the identification of suspected compounds by LC-MS/ms analysis were obtained compounds of germcarone, dehydrocurdione, camphene, isocurcumenol, and benzopyran with properties as antioxidant.
