UJI AKTIVITAS ANTIIHIPERLIPIDEMIA EKSTRAK ETANOL DAUN PELAWAN (Tristaniopsis merguensis Griff.) TERHADAP TIKUS JANTAN GALUR WISTAR YANG DIINDUKSI SUPLEMEN TINGGI LEMAK

Pelawan leaves (Tristaniopsis merguensis Griff.) are a plant that contain important secondary metabolites such as flavonoids, tannins, saponins, and phenolic compounds, which have potential roles in regulating lipid metabolism. This study aimed to evaluate the effect and effective dose of the ethanolic extract of pelawan leaves in reducing total cholesterol, Low Density Lipoprotein (LDL), and triglyceride levels, as well as increasing High Density Lipoprotein (HDL) levels in male Wistar strain white rats. A total of 30 rats were divided into five groups: a normal group, a negative control group (0.5% Na-CMC), a positive control group (simvastatin 0.9 mg/kg body weight), and three treatment groups receiving the extract at doses of 200, 300, and 400 mg/kg body weight. The rats were induced with propylthiouracil (PTU) and a high-fat diet for 30 days, followed by treatment for 14 days. Blood lipid parameters were measured using enzymatic colorimetric methods, namely Glycerol-3-Phosphate Oxidase–Para-Aminoantipyrine (GPO-PAP) for triglycerides, Cholesterol Oxidase–Para-Aminoantipyrine (CHOD-PAP) for total cholesterol, the precipitation method for HDL, and the indirect method for LDL. The results showed that the percentage reduction in total cholesterol levels (%PKKT) at doses of 200, 300, and 400 mg/kg body weight were 46.50%, 53.62%, and 57.65%, respectively, with the best effect observed at the dose of 400 mg/kg body weight. The relationship between the dose of the ethanolic extract of pelawan leaves and the percentage reduction of hyperlipidemia parameters indicated that the Effective Dose 50% (ED₅₀) value obtained was 204.6 mg/kg body weight. The dose of 400 mg/kg body weight demonstrated increasingly optimal antihyperlipidemic effects, and the macroscopic appearance of the liver was nearly comparable to that of the positive control group (p > 0.05). These findings indicate the potential of pelawan leaf ethanolic extract as a natural therapy for hyperlipidemia. Keywords : High-fat diet, hyperlipidemia, pelawan, propylthiouracil, simvastatin
