PENGARUH EKSTRAK ETANOL BUAH LEUNCA (Solanum nigrum. L) TERHADAP PERTUMBUHAN DAN AKTIVITAS PROBIOTIK Lactococcus lactis DALAM MENGHAMBAT PERTUMBUHAN Escherichia coli

This study was conducted to evaluate the activity of leunca fruit extract (Solanum nigrum Linn.), which contains flavonoid compounds, on the growth of the probiotic Lactococcus lactis. The combination of leunca fruit extract and L. lactis, each producing secondary metabolites, is expected to enhance the inhibition of Escherichia coli. Antibacterial activity was assessed by measuring the inhibition zones using the disc diffusion method. In addition, the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of the probiotic supplemented with the extract were determined. The characterization of leunca fruit extract showed the following results, specific gravity (0.12 ± 0.06), moisture content (5.66 ± 1.15), loss on drying (6.66 ± 0.57), water-soluble extractive content (37.6 ± 3.01), ethanol-soluble extractive content (39 ± 0.82), and total ash content (2.3 ± 0.57). Probiotic growth was evaluated using the Total Plate Count (TPC) method with four extract concentration variations (5%, 10%, 15%, and 20%). The highest probiotic growth was observed at the 20% extract concentration, with a colony count of 4.1 × 10⁸ CFU/mL. Each group was tested at concentrations of 40%, 65%, 80%, and 100%, along with positive and negative controls. The best antibacterial activity was observed in the group combining L. lactis metabolites and leunca fruit extract at 100% concentration, which produced an inhibition zone diameter of 17.83 ± 0.29 mm, categorized as strong inhibition. The KHM value obtained was a concentration of 3% in L. lactis metabolites combining leunca extract and single leunca extract, whereas in L. lactis metabolites it was 3.5%. The KBM value obtained was a concentration of 4.5% in L. lactis metabolites combining leunca extract and single leunca extract, and in L. lactis metabolites at a concentration of 5%.
