PENGARUH PEMBERIAN NAPHTALENE ACETIC ACID DAN BENZYL AMINO PURINE TERHADAP EKSPLAN DAUN KOPI ROBUSTA (Coffea canephora) PADA MEDIA MURASHIGE AND SKOOG
Robusta coffee can grow well in areas with an average annual temperature between 22ºC and 26ºC. This study aims to determine the effect and effectiveness of NAA (Naphthalene Acetic Acid) and BAP (Benzyl Amino Purine) on Robusta coffee leaf explants (Coffea canephora) using MS (Murashige and Skoog) medium. This study was conducted using a combination of NAA (Naphthalene Acetic Acid) and BAP (Benzyl Amino Purine). The NAA concentration used consisted of two levels, namely 0.5 ppm (N1) and 1 ppm (N2), while the BAP concentration consisted of three levels, namely 0 ppm (B0), 1 ppm (B1), and 2 ppm (B2). The results showed that treatment with 0.5 ppm NAA + 2 ppm BAP was the most effective in maintaining explant viability for 8 weeks after inoculation. This treatment also showed the best ability to induce callus formation, with a percentage of 56.3%. However, the best callus morphology quality was obtained in the treatment with 1 ppm NAA + 2 ppm BAP, which was characterized by a fresh green color and compact callus texture. This treatment also showed the lowest level of contamination. The results of this study can be used as a reference in determining the appropriate combination of NAA and BAP in robusta coffee tissue culture, particularly to achieve a balance between explant viability, callus formation, and callus morphological quality.
