KARAKTERISASI DAN UJI TOKSISITAS AKUT EKSTRAK ETANOL DAUN SUNGKAI (Peronema canescens Jack) PADA TIKUS WISTAR DENGAN FIXED DOSE PROCEDURE

Sungkai leaves (Peronema canescens Jack) possess various biological activities, including immunostimulatory, antidiabetic, antihyperuricemic, antibacterial, and antiparasitic effects. The leaves contain active compounds such as flavonoids, terpenoids, steroids, alkaloids, and phenolic compounds. This study aimed to evaluate the safety of the extract through toxicity testing and to perform extract standardization to ensure the quality of the test material. Organoleptic characterization showed that the extract was thick, greenish-black in color, had a characteristic odor, and a bitter-astringent taste. The results of physicochemical evaluation showed a moisture content of 6%, loss on drying of 6.39%, water-soluble extractive value of 66%, ethanol-soluble extractive value of 68.67%, total ash content of 5%, and acid-insoluble ash content of 0.2%, all of which met the requirements of the Indonesian Herbal Pharmacopoeia. Acute toxicity testing was conducted using the Fixed Dose Procedure, with a preliminary test at doses of 5, 50, 300, and 2000 mg/kg body weight, which showed no signs of toxicity or mortality. Therefore, a dose of 2000 mg/kg body weight was selected for the main test. In the main study, no mortality or behavioral changes indicative of toxic effects were observed. The extract did not cause significant changes in blood biochemical parameters. Macroscopic and histopathological examinations revealed normal liver morphology and structure in both groups.
