AKTIVITAS ANTIOKSIDAN DAN ANTIBAKTERI SERTA IDENTIFIKASI FRAKSI KROMATOGRAFI KOLOM EKSTRAK METANOL RIMPANG TEMU IRENG (Curcuma aeruginosa Roxb.)
Temu ireng (Curcuma aeruginosa Roxb.) is a medicinal plant from the Zingiberaceae family that is widespread in South Asia and Southeast Asia and has been reported to have antibacterial, anticancer, and antioxidant activities, especially in the rhizome. However, information on the antioxidant and antibacterial activities of fraction and isolated compounds is still limited. This study aims to assay the antioxidant and antibacterial activities of extracts and fractions column chromatography from black turmeric rhizome methanol extracts, then identify the chemical compounds contained in selected fractions of rhizome methanol extracts. Extraction was performed by maceration using methanol, followed by column chromatography fractionation and TLC analysis. Antioxidant assay was performed using the DPPH method and antibacterial assay against Escherichia coli and Staphylococcus aureus. The methanol extract and fraction F5 showed weak antioxidant activity with IC₅₀ values of 2071 µg/mL and 2077 µg/mL, respectively. Antibacterial activity of methanol extracts and F5 compared to tetracycline against E. coli was 31.77% and 43.61%, respectively, and against S. aureus was 31.71% and 33.65%, respectively. The antibacterial activity was classified as weak, with fraction F5B showing a MIC of 250 µg/mL against S. aureus. LC-MS/MS and FTIR analysis indicated the presence of procurcumenol, isocurcumenol, and curcumenol, supported by the identification of characteristic sesquiterpenoid functional groups.
