POTENSI EKSTRAK DAUN SIRIH HUTAN (Piper aduncum L.) SEBAGAI SKIN WHITENING AGENT MELALUI UJI AKTIVITAS ANTIOKSIDAN, INHIBISI ENZIM TIROSINASE, DAN NILAI SPF
Piper aduncum L. (sirih hutan) is a tropical plant widely distributed in Indonesia and has been empirically used in various traditional medicinal practices. The leaves of Piper aduncum have been reported to contain bioactive compounds such as phenolics, flavonoids, and terpenoids, which possess potential protective effects on the skin against oxidative stress, ultraviolet radiation, and pigmentation disorders. This study aimed to evaluate the potential of P. aduncum leaf ethanol extract as a skin whitening agent through antioxidant activity, tyrosinase inhibition, sun protection factor (SPF) value, and compound identification using LC-HRMS. Extraction was performed using the ultrasound sssisted extraction (UAE) method with ethanol as the solvent. Antioxidant activity was evaluated using the 2,2-diphenyl-1-pycrylhydrazyl (DPPH) method and expressed as IC₅₀ values. Tyrosinase inhibitory activity was assessed in vitro using L-DOPA as the substrate and expressed as IC₅₀ values, while SPF values were determined using the mansur method.The results showed that 177 compounds were identified in the ethanol extract, with phenolic compounds predominating (24.3%), along with the presence of flavonoids and terpenoids. The extract exhibited very strong antioxidant activity, with an IC₅₀ value of 42.84 µg/mL; however, tyrosinase inhibitory activity was very weak, with an IC₅₀ value of 13,180.28 µg/mL. The SPF values ranged from 3.71 to 11.78, indicating minimal to high photoprotective activity. Based on these findings, the ethanol extract of P. aduncum leaves shows potential as a skin whitening agent primarily through antioxidant and photoprotective mechanisms. However, enhancement of tyrosinase inhibitory activity may require further development, such as fractionation or isolation of active compounds.
