AKTIVITAS BIOLOGIS DAN IDENTIFIKASI TENTATIF SENYAWA PADA FRAKSI KOLOM EKSTRAK METANOL RIMPANG TEMU MANGGA (Curcuma amada Roxb.)
ABSTRACT : Increasing antibiotic resistance and oxidative stress in various diseases encourage the search for sources of natural bioactive compounds. Mango ginger (Curcuma amada Roxb.) is a plant from the Zingiberaceae family known to contain secondary metabolites with potential biological activity. This study aims to test the antioxidant and antibacterial activities and tentatively identify compounds in column chromatography fractions of methanol extract of mango ginger rhizome. Extraction was carried out by maceration method using methanol, followed by separation of fractions using gravity column chromatography and TLC analysis. Antioxidant activity was tested by the DPPH method, while antibacterial activity was determined by the disc diffusion method and determination of the minimum inhibitory concentration against Escherichia coli and Staphylococcus aureus. Active fractions were analyzed using LC-MS/MS and FTIR. The results showed that the methanol extract and F2 fraction had weak antioxidant activity with IC50 values of 1456 µg/mL and 1986 µg/mL, respectively. Subfraction F2.2 showed the highest antibacterial activity with MIC values against E. coli of 0.9765 µg/mL and S. aureus of 3.90625 µg/mL, and inhibition zone diameters of 7.5 mm and 7.25 mm. LC-MS/MS and FTIR analysis showed that subfraction F2.2 was suspected to contain tentative compounds (E)-labda-8(17),12-dien-15,16-dial and 12β hydroxy-15-norlabda-8(17),13(14)-dien-16-oic acid which are labdane diterpenoids.
