PENGARUH PEMBERIAN EKSTRAK ETANOL SIPUT GONGGONG (LAEVISTROMBUS SP.) TERHADAP PERUBAHAN PARAMETER HISTOLOGIS DAN KADAR INTERLEUKIN-10 PADA PROSES PENYEMBUHAN LUKA EKSISI TIKUS PUTIH

Introduction: Wound healing is a complex biological process involving the inflammatory, proliferative, and tissue remodeling phases. Interleukin-10 (IL-10) acts as an anti-inflammatory cytokine that supports tissue regeneration during wound healing. Gonggong snails (Laevistrombus sp.) contain antimicrobial peptides (AMPs), collagen, and bioactive compounds such as flavonoids, alkaloids, and saponins, which have potential anti-inflammatory properties. The SPOT score is a histological parameter used to evaluate epithelialization, keratinization, epithelial thickness index, granulation tissue formation, scar elevation index, and tissue remodeling during the wound healing process.This study aimed to determine the effect of ethanol extract of gonggong snails on SPOT score and IL-10 levels during excision wound healing in white rats. Methods: Laboratory experimental research used 30 Wistar strain white rats was conducted with a post-test only control group design, involving a negative control group, a positive control group, and treatment groups with 5%, 10%, and 15% concentration extracts administered topically. Evaluation was carried out on days 7 and 14 through histological assessment using the SPOT score and immunohistochemical examination of IL-10 expression. Data were analyzed using one way ANOVA and post hoc tests. Result: On day 7, a significant increase in SPOT score was observed in the 15% extract group compared with the negative control (p=0.005). On day 14, higher SPOT scores were observed in the 5% extract group (p=0.035), 10% extract group (p=0.043), and 15% extract group (p=0.034) compared with the negative control. IL-10 levels showed no significant difference on day 7 (p=0.520). However, on day 14, IL-10 levels increase in the 5% extract group (IRS=7), although this increase was not statistically significant (p=0.657). Conclusion: The ethanol extract of gonggong snail affected the SPOT score and increased IL-10 levels in the 5% extract group; however, the increase in IL-10 levels was not statistically significant.
