AKTIVITAS ANTIOKSIDAN SENYAWA METABOLIT SEKUNDER DAUN SEMBUNG RAMBAT (Mikania micrantha Kunth)
Free radicals are reactive molecules that can damage cells and trigger various degenerative diseases when present in excessive amounts. Antioxidants play an important role in neutralizing free radicals, and medicinal plants are recognized as sources of natural antioxidants that are relatively safer than synthetic antioxidants. One plant with potential as a source of antioxidants is Mikania micrantha (sembung rambat), which contains a variety of bioactive secondary metabolites. However, research on the isolation and antioxidant activity of pure compounds from the leaves of sembung rambat remains limited. This study aims to identify antioxidant compounds present in the n-hexane, ethyl acetate, and methanol–water fractions of sembung rambat leaves, determine their compound classes, and evaluate the IC₅₀ values of the obtained isolates. The research was conducted from August to December 2025 at the Genetics and Biotechnology Laboratory also Physiology and Development Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University. The methods employed included extraction, fractionation, thin-layer chromatography (TLC) analysis, isolation using vacuum liquid chromatography and column chromatography, DPPH assay using a UV-Vis spectrophotometer, and IC₅₀ value analysis. The results showed that the n-hexane and ethyl acetate fractions contained antioxidant compounds, whereas the methanol–water fraction did not. Five pure isolates (N1, N2, N3, E1 and E2) were obtained. The identified compounds belong to the steroid group (N1, N3), terpenoid group (N2, E2) and flavonoid group (E1) with IC₅₀ values of 88.82 ppm, 113.48 ppm, 76.31 ppm, 82.57 ppm and 49.06 ppm.
