UJI AKTIVITAS IMUNOSTIMULAN EKSTRAK ETANOL DAUN PEPAYA (Carica papaya L.) TERHADAP TIKUS PUTIH JANTAN GALUR WISTAR DENGAN METODE CARBON CLEARANCE
The papaya leaf (Carica papaya L.) is known to contain various bioactive compounds, particularly flavonoids, which have potential Immunostimulatory activity. This study aimed to evaluate the Immunostimulatory effect of the 70% ethanolic extract of papaya leaves using the carbon clearance method in male Wistar rats. The experimental animals were divided into six groups, consisting of a normal control, a negative control (NaCMC 0.5% + carbon), a positive control (Stimuno® 150 mg/kg BW + carbon), and three treatment groups administered papaya leaf ethanolic extract at doses of 200, 400, and 800 mg/kg BW. The observed parameters included phagocytic index, phagocytic constant, total leukocyte count, and lymphocyte percentage. The results showed that administration of papaya leaf ethanolic extract significantly increased phagocytic activity, phagocytic index, total leukocytes, and lymphocyte percentage compared to the negative control group (p < 0.05). The increase in immunological parameters tended to be dose-dependent, with the highest effect observed at a dose of 800 mg/kg BW. Statistical analysis using One Way ANOVA followed by Duncan’s Multiple Range Test confirmed significant differences among treatment groups. In conclusion, the 70% ethanolic extract of papaya leaves exhibits Immunostimulatory activity by enhancing phagocytic function and regulating cellular immune responses in male Wistar rats induced with carbon. These findings support the potential use of papaya leaves as a natural Immunostimulatory agent. Keywords: Carbon clearance, Carica papaya L., immunostimulator, leukocytes, phagocytosis.
