POTENSI DAN KARAKTERISASI KANDUNGAN KOLAGEN Holothuria atra, Culcita novaeguineae, dan Glauconome virens DI PERAIRAN PULAU SEBESI, LAMPUNG SELATAN

Holothuria atra, Culcita novaeguineae, and Glauconome virens are marine invertebrates that have potential as sources of collagen and bioactive compounds, including natural antioxidants. The utilization of collagen from marine organisms serves as an alternative to collagen derived from terrestrial animals, which has limitations in terms of religious concerns and disease risks. This study aimed to analyze the yield and characterize collagen from H. atra, C. novaeguineae, and G. virens collected from the waters of Sebesi Island, South Lampung. The research method involved purposive sampling in the waters of Sebesi Island, Lampung Province. Collagen isolation was carried out using the acid soluble collagen (ASC) method, which produced collagen yields of 0.35% (H. atra), 0.28% (C. novaeguineae), and 0.24% (G. virens). Antioxidant activity testing using the DPPH method showed IC₅₀ values of 1130.68 µg/mL (H. atra), 380.175 µg/mL (C. novaeguineae), and 1267.36 µg/mL (G. virens), which are categorized as very weak antioxidant activity. Functional group characterization using FTIR showed the presence of Amide A, B, I, II, and III. LC-MS analysis of collagen from C. novaeguineae identified various amino acids composing collagen, namely glutamic acid, aspartic acid, leucine, isoleucine, alanine, valine, lysine, threonine, glycine, serine, proline, methionine, phenylalanine, histidine, and arginine. The results of this study indicate that H. atra, C. novaeguineae and G. virens from the waters of Sebesi Island have potential as a source of marine collagen, although the antioxidant activity of the isolated collagen is still relatively low.
