DNA BARCODE IKAN LUNDU (Mystus gulio) ASAL SUNGAI MUSI DAN SUNGAI BATANGHARI BERDASARKAN GEN SITOKROM C OKSIDASE SUBUNIT I (COI)
Long whiskers catfish (Mystus gulio) is fish species from the Bagridae family of the Mystus genus. Long whiskers catfish in Indonesia were found in Java, Kalimantan and Sumatra. This research aimed to determine the COI gene sequence of mitochondrial DNA, the genetic distance and phylogenetic of long whiskers catfish habitat. This research was carried out in December 2023-March 2024. Fish samples (n= 15) and water samples were taken from the Musi River, Batanghari River and downstream of the Batanghari River. The stages of DNA barcoding included DNA extraction, DNA amplification using polymerase chain reaction (PCR), electrophoresis and COI gene sequencing. The COI mtDNA gene fragment was amplified at an annealing temperature of 50℃ for 30 seconds in 30 cycles (samples from the Musi River and downstream of the Batanghari River) and 51.5℃ for 30 seconds in 30 cycles (samples from the Batanghari River). The nucliotide length of the COI gene from Musi River and downstream of the Batanghari River was 651 bp, while from the Batanghari River was 646 bp. BLASTn analysis indicated the samples from Musi River and downstream of the Batanghari River had the highest similarity of 98.40% to the Mystus gulio meanwhile samples from the Batanghari River had the highest similarity of 98.41% to the Mystus sp. Phylogenetic analysis indicated that the long whiskers catfish from Musi River and downstream of the Batanghari River were in a separate cluster from Batanghari River. The results of the measurement of water quality from three locations were temperature of 29.2-30.5°C, pH 7-7.10, dissolved oxygen 4.1-6.5 mg L-1, TDS 0.02-0.04 mg L-1, transparency 32-42 cm, ammonia 0.09-0.2 mg L-1 and alkalinity 26-45 mg L-1.
